When autoclaving microbial culture media, moisture-proof paper or kraft paper should be used to wrap the cotton plugs of test tubes, with the tube opening facing upwards, to prevent the cotton plugs from becoming damp during sterilization. Kraft paper is used because it is effectively waterproof, ensuring the test tubes do not get wet.
